They will hire you but you mite have to go to training doing miles with a trainer. For example I worked for Prime did 40000 with a trainer got my own truck worked for them for some time, Then went local now I want to go otr again at first they were going to have me go 15000 miles with a trainer. But my old FM talked to the safety department and now I don't have to.
OTR driving normally means you'll be hauling freight to various customers throughout your company's hauling region. It often entails being gone from home for two to three weeks at a time.
A lot of companies just specify tractor trailer experience, pup would count. Depends on the company.

I'm going to be driving a truck with a pup trailer making local deliveries. Let's say in a year or two I wanna do regional or OTR would the companies still hire me?
Regional:
Regional Route
Usually refers to a driver hauling freight within one particular region of the country. You might be in the "Southeast Regional Division" or "Midwest Regional". Regional route drivers often get home on the weekends which is one of the main appeals for this type of route.
OTR:
Over The Road
OTR driving normally means you'll be hauling freight to various customers throughout your company's hauling region. It often entails being gone from home for two to three weeks at a time.
