They'll work with you as much as you can. I run two weeks out and home on the weekends I have my daughter. My DM gets me home Friday evening and I generally leave out late Sunday night or early Monday morning. Getting a weekly route right off the bat won't be that easy to do but it depends what terminal you're out of.
A facility where trucking companies operate out of, or their "home base" if you will. A lot of major companies have multiple terminals around the country which usually consist of the main office building, a drop lot for trailers, and sometimes a repair shop and wash facilities.

They do some lifting exercises, some stair step too. They want to elevate your heart rate and make sure it comes back to a good rating heart rate in a reasonable time frame. You'll also simulate pulling a 5th wheel release and a couple other simple things. It's mainly a way to make sure beforehand that you're capable of the day to day duties that go along with the work. I wouldn't sweat it too much.

Regional:
Regional Route
Usually refers to a driver hauling freight within one particular region of the country. You might be in the "Southeast Regional Division" or "Midwest Regional". Regional route drivers often get home on the weekends which is one of the main appeals for this type of route.
OTR:
Over The Road
OTR driving normally means you'll be hauling freight to various customers throughout your company's hauling region. It often entails being gone from home for two to three weeks at a time.
