No, as long as your schooling and CDL driving exam was done in a truck with a standard transmission. If you took the test in an automatic you would have a restriction on your license preventing you from driving standard transmissions. But as long as you have no restrictions you'll be fine. You'll have to take a driving test anytime you start a new job - that's a Federal requirement actually. So if you're really struggling to shift they may have you take some time on the road with a trainer or practicing around the yard or something. But you'll be able to find work.
