Prime has classes starting every Monday in both Springfield MO and SLC Utah. I applied over a weekend, got my email from them on Monday...returned all their info by Wed and was on a bus THAT Sat. So seriously it was a week. However, they will ask you when you need to come...give notice to your employer etc.
As for the written test...you get to take the four sections twice a day from Tues -Fri. If you don't pass after that, you get sent home. If you follow the High Road you WILL pass.
TWIC:

Your TWIC is a tamper-resistant biometric card which acts as both your identification in secure areas, as well as an indicator of you having passed the necessary security clearance. I'm organized and had all docs readily available. Also I have no issues with my past or DMV record. Same job for almost two decades. So as far as they were concerned I was a gem.

Three years ago I applied to Swift and two weeks later I started CDL training. You might get in faster than that. On the other side, you can look for starting four weeks away. The recruiter will make sure all the boxes are checked - that all your documents are in, and they can work with you on the timing.
Also, some training classes might be booked up for the next month or two. Its best to to be talking with your recruiters sooner rather than later.
